Cellulase production from lignocellulosic material by Trichoderma reesei

In general, cellulase is produced on pure cellulose. Very little research has been done to use crude sources of cellulose such as wood. The present study deals with lignocellulosic material (Chemithermomechanical pulp: CTMP) as a substitute for pure cellulose. The cellulase produced by growing mutant Trichoderma reesei QMy-1 in a CTMP solution gave filter paper activity (FPA) of 2.5 IU/ml at an optimal pH of 6. The FPA yield was 416 IU/g cellulase, and the cellulase conversion rate was 3 IU/g cellulase/h. A two-phase enzyme production was observed: in the first phase, amorphous cellulose and hemicelluloses were consumed; in the second phase, the crystalline portion of cellulose was used to produce cellulase. A drop of about 20% in FPA was observed during the shift from the first phase to the second.
