The operational normality of an enzyme solution can be determined by observation of a specific binding equilibrium, up to conditions of binding site saturation. For enzymes with one site, titrations are given by either eq. 1 (E >> K), or by eqs. 2 and 3 (E ≃ K). In eqs. 1-3, A is a property of the system proportional to the concentration of enzyme-reagent complexes, with proportionality constant Σ; A(max) is the value of A at saturation; K is the binding (dissociation) constant; E and S are the total concentrations of enzyme and reagent, respectively.
