Mass spectrometry, often combined with chromatography, has evolved as an important technique for the analysis of structural modification in nucleic acids. These modifications serve a range of biological purposes, and occur over a wide frequency range in both RNA and DNA. Transfer RNA (tRNA) constitutes the most remarkable example, in which more than 55 nucleosides have thus far been discovered. A substantial analytical challenge results from limitations imposed by the high polarity of nucleosides and nucleotides, and the necessity of working with limited quantities of material often present in complex mixtures. The development of combined liquid chromatography/mass spectrometry, and of fast atom bombardment and related techniques, have added an important dimension to the design of microscale procedures applicable to RNA or DNA.
