Towards nanostructured digital microfluidic-based SPR biosensors

The development of biosensors for point-of-care diagnosis requires on- field analysis accompanied by real-time data acquisition. While SPR imaging (SPRi) biosensors could potentially be the best suited candidate for biomarker-based diagnosis, there remains many challenges to overcome. In particular, they are limited by the flow-cell technology to serial-sample processing and have limited sensitivity for the detection of markers present at low concentration. In this work, we explored SPRi technology and implemented an approach to increase sensitivity and enhance surface plasmon resonance imaging detection of DNA binding interactions. We have developed a droplet-based digital microfluidic platform with an integrated nanostructured biosensor interface that allows for ultra-low volume, sensitive, two-dimensional, and automated SPRi detection of DNA hybridization reactions. The limit of detection was estimated to be 500 pM (90 attomoles) in 180 nL droplets, allowing rapid DNA target identification within only 60 seconds of the reaction. Coupled with SPRi, nanostructured EWOD biochip platforms promise to dramatically increase the sensitivity and speed of biomolecule’s detection beyond DNA biosensors. The proposed platform allows extreme versatility in the manipulation of precious samples which makes this technology very suitable for point of care diagnosis.
