Induction of ovulation in adult female rats by various combinations of PMS and HCG

Ovulation was induced in adult female rats by timely injected PMS followed by HCG. Autopsy was done 72 hrs following PMS injection and the tubal eggs were counted. PMS or HCG alone was ineffective in inducing ovulation but PMS at a dose of as low as 10 i.u. would readily induce ovulation if followed by 10 i.u. HCG (9.4 ± 1.0 eggs/rat). Increased number of ova was obtained with higher doses of PMS (30 i.u. or more) followed by 10 i.u. or more of HCG (12.9 ± 2.0 or more eggs/rat). Ovulation was induced in 24-day old immature Sprague Dawley female rats by timely injected 30 i.u. pregnant mare serum gonadotrophin (PMS). Administration of progesterone, norethynodrel, Enovid, lynestrenol and medroxy-progesterone acetate 48-52 hrs following PMS injection increased the number of eggs/ovulating rat as well as the % rats ovulated. Estrogens and androgens showed no such facilitatory effect on PMS induced ovulation.

Adult female mice were injected with a norethynodrel (98.5%) and mestranol (1.5%) combination (EnovidR) at doses of 200 ug. per mouse per day and 50 ug. per mouse per day for 15 days. Organ weights at autopsy showed increased uterine weight and decreased ovarian weight; and are suggestive of increased pituitary weight and decreased adrenal weight. The drug decreased the rate of body weight gain in the mice. Preliminary growth hormone content determination of the pituitaries showed no changes.
