Mass spectrometry, often combined with chromatography, has evolved as an important technique for the analysis of structural modification in nucleic acids. These modifications serve a range of biological purposes, and occur over a wide frequency range in both RNA and DNA. Transfer RNA (tRNA) constitutes the most remarkable example, in which more than 55 nucleosides have thus far been discovered. A substantial analytical challenge results from limitations imposed by the high polarity of nucleosides and nucleotides, and the necessity of working with limited quantities of material often present in complex mixtures.
